Pregnenolone

What is Pregnenolone?

Pregnenolone is a steroid hormone that serves as a precursor to other hormones, including testosterone, DHEA, estrogen, and cortisol. It is primarily produced in the adrenal glands, with smaller amounts generated in the skin, brain, gonads, liver, and eyes.

In the body's hormone cascade, pregnenolone is at the top, synthesized directly from cholesterol. This connection suggests that excessively low cholesterol levels can contribute to hormone imbalances. Low levels of pregnenolone are linked to deficiencies in other more active hormones.

Pregnenolone is also concentrated in the brain, where it supports cognitive health and overall brain function. It helps produce neurohormones that maintain nerve cell function and regulate mood. As pregnenolone levels decline with age—sometimes by as much as 60%—researchers have associated low levels with depression, anxiety, cognitive deficits, and other mental health issues in older adults.

What Does Pregnenolone Do for Men?

Besides acting as the main precursor for most steroid hormones, pregnenolone has a number of important functions in the body. Low levels of pregnenolone have been linked to memory problems and cognitive deficits, fatigue and low energy, low mood, and even increased symptoms of menstruation.

Unfortunately, researchers haven’t focused on the specific effects of pregnenolone in the body. This has more to do with how drugs are researched than it has to do with pregnenolone. In the 1940s, when researchers were just beginning to unravel how adrenal hormones worked, pregnenolone was extensively studied alongside cortisol for the relief of inflammatory conditions such as arthritis. In the 1950s, however, researchers figured out how to make synthetic cortisol, so the research focused on the more profitable hormone and the study of pregnenolone by itself declined.

Low Pregnenolone Symptoms in Men

As the primary precursor for steroid hormones, pregnenolone is often part of a comprehensive hormone panel. Low levels typically indicate an overall hormone imbalance, but isolating specific symptoms related to low pregnenolone can be challenging, as they may overlap with the effects of more active hormones like testosterone.

Symptoms of low pregnenolone in men may include:

- Hot flashes
- Poor memory
- Low mood
- Irritability
- Anxiety
- Reduced levels of neurohormones
- Decreased libido
- Low energy
- Fatigue
- Limited mobility
- Impaired brain function

Pregnenolone Benefits for Men

Bioidentical hormone replacement programs often use pregnenolone as a balancing hormone. Evidence suggests that maintaining healthy pregnenolone levels can support overall hormone balance, boost the immune system, reduce stress, and potentially prolong life. Pregnenolone is commonly used alongside testosterone and estrogen to alleviate symptoms of estrogen imbalance, andropause, and testosterone deficiency in men.

Benefits of pregnenolone supplementation may include:

- Improved memory
- Enhanced mood and relief from depression
- Reduced anxiety
- Increased levels of neurohormones
- Alleviation of low testosterone symptoms (e.g., reduced libido, low energy)
- Relief from fatigue
- Better mobility
- Improved brain function

The hormone produced from pregnenolone varies based on where it is synthesized. For example, pregnenolone from the adrenal glands typically becomes cortisone, while pregnenolone from the ovaries is likely to be converted into estrogen. The goal of pregnenolone supplementation is to support overall healthy levels, encouraging the body to produce the necessary steroid hormones where needed. Additionally, pregnenolone can be applied topically, with some evidence suggesting it may reduce wrinkles and improve skin elasticity.
